Studies on the Use of Rabbits in Evaluation of Drug Bioavailability with Special Reference to Prednisolone






Abstract
To investigate the possibility for the use of rabbits in evaluation of drub bioavailability tire effect of feed types (soft and solid) and coprophagy prevention on the amount of feces execreted and remaining stomach contents were examined. In addition, serum prednisolone concentrations according to presence or absence and types of gastric contents were measured.
The results obtained were summarized as follows;
1. Amounts of feces execreted were significantly larger(p$lt;0.001) in groups with cangues (soft diet; 40, 50¡¾2.64g) solid diet; 37, 78¡¾7.57g) than in those without cangues (soft diet; 8.04¡¾1.27g, soil diet; 8.92¡¾2.43g), and no statistical differences between feed types (soft and solid) were observed.
2. Amounts of dried gastric contents were significantly larger(p$lt;0.001) in group fed solid diet without cangues(16.06¡¾1.25g) than in that fed soft diet with cangues(2.24¡¾0.83g)
3. Peak serum level of prednisolone was higher (p$lt;0.05) in the stomach-emptying-controlled group administered drug with water(4.931¡¾0.932ug/§¢) than in the conventionally fasted group(2.789¡¾0.233ug/§¢).
4. Time to reach maximal serum prednisolone level was shorter (p$lt;0.01) in the stomachemptying-controlled group administered drug with water (1.1¡¾0.15hr) than in the conventionally fasted group (1.7¡¾0.12hr).
5. There were no statistical differences in the area under the serum concentration-time curves among the four groups.
6. The pattern of serum prednisolone concentration-time curves in the stomach-emptying-controlled rabbits was similar to that in humans.
7. The laboratory animal feed marketed in Korea was satisfactory to maintain the stomach-emptying-controlled rabbits.
